Metaproteogenomic Profiling of Chemosynthetic Microbial Biofilms Reveals Metabolic Flexibility During Colonization of a Shallow-Water Gas Vent

چکیده

Tor Caldara is a shallow-water gas vent located in the Mediterranean Sea, with active venting of CO 2 and H S. At Caldara, filamentous microbial biofilms, mainly composed Epsilon- Gammaproteobacteria , grow on substrates exposed to venting. In this study, we took metaproteogenomic approach identify metabolic potential situ expression central pathways at two stages biofilm maturation. Our findings indicate that inorganic reduced sulfur species are main electron donors carbon source for which conserve energy by oxygen nitrate respiration, fix dinitrogen detoxify heavy metals. Three metagenome-assembled genomes (MAGs), representative key members community, were also recovered. Metaproteomic data show metabolically chemoautotrophic sulfide-oxidizing Epsilonproteobacteria dominated young while become prevalent established community. The co-expression different sulfide oxidation these classes bacteria suggests exposure concentrations within as well fine-tuned adaptations enzymatic complexes. Taken together, our demonstrate shift taxonomic composition associated activity biofilms course colonization process.

Galenea microaerophila gen. nov., sp. nov., a mesophilic, microaerophilic, chemosynthetic, thiosulfate-oxidizing bacterium isolated from a shallow-water hydrothermal vent.

A mesophilic, strictly microaerophilic, chemosynthetic bacterium, designated strain P2D(T), was isolated from the sediment of an active shallow-water hydrothermal vent in Paleochori Bay, on the Greek island of Milos. The cells were Gram-staining-negative rods that measured approximately 0.8-1.3 µm in length and 0.4-0.5 µm in width. Strain P2D(T) grew at 20-50 °C (optimum 35 °C), with 1.0-5.0% (...

Metabolic Profiling as a Screening Tool for Cytotoxic Compounds: Identification of 3-Alkyl Pyridine Alkaloids from Sponges Collected at a Shallow Water Hydrothermal Vent Site North of Iceland

Twenty-eight sponge specimens were collected at a shallow water hydrothermal vent site north of Iceland. Extracts were prepared and tested in vitro for cytotoxic activity, and eight of them were shown to be cytotoxic. A mass spectrometry (MS)-based metabolomics approach was used to determine the chemical composition of the extracts. This analysis highlighted clear differences in the metabolomes...

Microbial Colonization of Laparoscopic Gas Delivery Systems: A Qualitative Analysis

OBJECTIVE Laparoscopic procedures utilize a pneumoperitoneum to distend and separate the abdominal wall from the intra-abdominal structures. Carbon dioxide is commonly used for this purpose, although this study is inclusive of any gas used for abdominal distention. The gas is delivered from cylinders through a gas insufflation delivery system. The purpose of this study is to determine if laparo...

Metagenome analysis of an extreme microbial symbiosis reveals eurythermal adaptation and metabolic flexibility.

Hydrothermal vent ecosystems support diverse life forms, many of which rely on symbiotic associations to perform functions integral to survival in these extreme physicochemical environments. Epsilonproteobacteria, found free-living and in intimate associations with vent invertebrates, are the predominant vent-associated microorganisms. The vent-associated polychaete worm, Alvinella pompejana, i...
